Thy1-yfp Mouse

The Thy1-YFP mouse is a transgenic reporter model in which yellow fluorescent protein (YFP) labels selected neurons, enabling researchers to visualize neuronal structure and development in intact tissues. In this line, regulatory elements from the Thy1 gene drive YFP expression in subsets of differentiated neurons, producing fluorescence in cell bodies, axons, dendrites, and synaptic processes. Developmental biologists use Thy1-YFP mice to follow neuronal migration, axon growth, dendritic remodeling, and circuit formation with fluorescence microscopy, including longitudinal or live imaging approaches. The model links cellular morphology to nervous-system development and helps assess structural changes in neurodevelopmental disease and injury.

Multiphoton Microscopy of Cleared Mouse Brain Expressing YFP

Multiphoton microscopy of whole mouse organs is possible by optically clearing the organ before imaging, but not all protocols preserve the fluorescent signal of fluorescent proteins. Using an optical clearing method with ethanol-based dehydration and benzyl alcohol:benzyl benzoate clearing, we show high-resolution multiphoton images of whole mouse brain expressing YFP.

Intravital Two-Photon Imaging of Touch Sensory Axon Morphology in Mouse Skin

This study presents an in vivo two-photon imaging approach for visualizing the structures of low-threshold mechanoreceptor (LTMR) axon terminals in the forepaw skin of mice. Using repeated, high-resolution imaging of individual axons, this method provides a new platform for studying the structure and function of the cutaneous sensory circuit during development and in adults.

Optical clearing techniques are revolutionizing the way tissues are visualized. In this report we describe modifications of the original Clear Lipid-exchanged Acrylamide-hybridized Rigid Imaging-compatible Tissue-hYdrogel (CLARITY) protocol that yields more consistent and less expensive results.
