Interrogator System

An interrogator system is an engineered instrument that supplies a stimulus to a sensor, measures its response, and converts that response into information about a physical condition. Depending on the sensor technology, it may generate optical, electrical, or acoustic signals, detect changes in wavelength, intensity, phase, frequency, or resistance, and process them through signal conditioning and data analysis. In engineering, interrogator systems enable distributed and multiplexed sensing for temperature, strain, pressure, vibration, and structural changes. They support real-time monitoring of infrastructure, machinery, and industrial processes, improving measurement accuracy, fault detection, and predictive maintenance.

This protocol outlines two methods for the quantitative analysis of mitophagy in pancreatic β-cells: first, a combination of cell-permeable mitochondria-specific dyes, and second, a genetically encoded mitophagy reporter. These two techniques are complementary and can be deployed based on specific needs, allowing for flexibility and precision in quantitatively addressing mitochondrial quality control.

A significant barrier to technologies like CRISPR is the off-target events that can disrupt vital genes. 'Circularization for In Vitro Reporting of Cleavage Effects by Sequencing' (CIRCLE-seq) is a technique designed to identify unintended cleavage sites. This method maps the genome-wide activity of CRISPR-Cas9 with high sensitivity and without bias.
