Sharp-edged Orifice

A sharp-edged orifice is a thin-plate opening with a deliberately abrupt, well-defined edge that restricts fluid flow and enables its measurement or control. As fluid passes through the aperture, the sudden contraction forms a vena contracta, where the stream reaches its minimum cross-sectional area; the resulting pressure drop and flow velocity depend on the opening diameter, upstream and downstream pressures, and fluid properties. Engineers use sharp-edged orifice plates in pipelines for flow-rate measurement, pressure reduction, and process control, often with differential-pressure instrumentation and discharge coefficients to improve accuracy. Their simple construction supports economical testing and monitoring, although permanent pressure loss and sensitivity to installation conditions must be considered.

The full process from brain specimen preparation to serial sectioning imaging using the Knife-Edge Scanning Microscope, to data visualization and analysis is described. This technique is currently used to acquire mouse brain data, but it is applicable to other organs, other species.
