Vascular Staining

Vascular staining is a set of histological methods used to visualize blood vessels or plant vascular tissues, making their structure and distribution measurable. In animal tissues, stains may be delivered through perfusion or applied to sections, where dyes or labeled probes bind vessel-associated structures and reveal features such as vessel walls, lumens, branching, and density; in plants, contrasting stains help distinguish xylem and phloem. Researchers use these approaches to compare vascular organization across tissues, species, and environmental conditions. The resulting images support studies of transport, development, tissue injury, and vascular responses to stress, linking microscopic structure with broader environmental and physiological effects.

Here, we present a protocol for immunofluorescence staining to observe the endothelial cells of the mouse aorta directly. This technique is useful when studying the cellular and molecular phenotype of endothelial cells in different flow patterns and in the development of atherosclerosis.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.

Assessment of Vascular Regeneration in the CNS Using the Mouse Retina

The rodent retina has long been recognized as an accessible window to the brain. In this technical paper we provide a protocol that employs the mouse model of oxygen-induced retinopathy to study the mechanisms that lead to failure of vascular regeneration within the central nervous system after ischemic injury. The described system can also be harnessed to explore strategies to promote regrowth of functional blood vessels within the retina and CNS.
