Absolute Qpcr

Absolute qPCR is a quantitative polymerase chain reaction method that determines the starting amount of a specific DNA target rather than reporting only relative changes, making it valuable for precise genetic measurements. During amplification, target-specific primers and fluorescent dyes or probes generate a signal that increases with each cycle, while the threshold cycle reflects the initial template concentration. Researchers compare this cycle value with a standard curve prepared from samples containing known target quantities to calculate absolute copy number or concentration. In genetics, absolute qPCR supports gene copy-number analysis, transgene validation, mutation detection, and measurement of nucleic acid abundance in biological samples.

Quantifying Environmental Microorganisms and Viruses Using qPCR

2023

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Bradley Schmitz Quantitative polymerase chain reaction (qPCR), also known as real-time PCR, is a widely-used molecular technique for enumerating microorganisms in the environment. Prior to this approach, quantifying microorganisms was limited largely to classical culture-based techniques. However, the culturing of microbes from environmental samples can be particularly challenging, and...

Merging Absolute and Relative Quantitative PCR Data to Quantify STAT3 Splice Variant Transcripts

Tandem splicing events occur at sites less than 12 nucleotides apart. Quantifying ratios of such splice variants is feasible using an absolute quantitative PCR approach. This manuscript describes how splice variants of the gene STAT3, in which two splicing events results in Serine-701 inclusion/exclusion and α/β C-termini, can be quantified.

Absolute Value

Absolute value describes the magnitude of a number without considering its direction. On a number line, positive numbers lie to the right of zero, while negative numbers lie to the left. Although two numbers may be on opposite sides of zero, they can be the same distance from the origin. For example, 2 and −2 are both two units from zero. The absolute value of a real number is its distance from zero on the number line. It is written using vertical bars. For example, |2| = 2 and |−2| = 2. Since...

Absolute Value Functions

The absolute value function is a fundamental mathematical concept that measures distance or magnitude without regard to direction, converting any real number or variable expression into a non-negative value. Denoted using vertical bars or the modulus symbol, the basic parent function evaluates inputs through a two-part rule: if an input is positive or zero, it remains unchanged; if an input is negative, it is multiplied by negative one to produce a positive output. The graph of an absolute value...
