Facs Enrichment

Fluorescence-activated cell sorting (FACS) enrichment is a method for increasing the proportion of cells with a desired phenotype by separating populations according to fluorescence and light-scattering properties, making it valuable in genetics and cell biology. Cells are labeled with fluorescent antibodies or express reporter proteins, then pass singly through a laser; detectors measure each cell, and an instrument directs selected droplets into separate containers through electrostatic deflection. Enriched populations can support DNA, RNA, and functional analyses, including studies of gene expression, mutant phenotypes, cellular heterogeneity, and rare genetic subpopulations, while improving the sensitivity and interpretability of downstream experiments.

Fibroblast behavior underlies a spectrum of clinical entities, but they remain poorly characterized, largely due to their inherent heterogeneity. Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.
