Genetic Tool Generation

Genetic tool generation is the design and development of molecular systems that enable researchers to modify, regulate, or measure genetic information, making it central to experimental genetics. It typically combines a chosen DNA sequence or regulatory element with a delivery and readout strategy, followed by testing in cells or organisms to determine whether the tool produces the intended genetic change or signal. These tools support gene editing, expression control, reporter assays, functional genomics, and disease modeling while improving the precision and interpretability of genetic experiments. Advances in tool generation can expand which genes and biological processes researchers can study.

The microinjection of mouse oocytes is commonly used for both classic transgenesis (i.e., the random integration of transgenes) and CRISPR-mediated gene targeting. This protocol reviews the latest developments in microinjection, with a particular emphasis on quality control and genotyping strategies.

Mendelian Genetics Evolution is caused by changes in the genetic composition of populations. In the field of population genetics, scientists model this process as changes in the frequency of alleles at individual genetic loci. This simple representation of how evolution occurs dates to Gregor Mendel’s analysis of trait inheritance patterns in pea plants, first presented in 1865. Mendel determined, using rigorous collection of data, that noticeable traits are controlled by two alleles of each...

2023

Genetic engineering – the process of purposefully altering an organism’s DNA – has been used to create powerful research tools and model organisms, and has also seen many agricultural applications. However, in order to engineer traits to tackle complex agricultural problems such as stress tolerance, or to realize the promise of gene therapy for treating human diseases, further advances in the field are still needed. Important considerations include the safe and efficient delivery of genetic...
