Parkinson's Disease Models

Parkinson’s disease models are experimental systems used to reproduce selected molecular, cellular, or organismal features of Parkinson’s disease and investigate its causes. They include genetically engineered animals, patient-derived cells, and other model platforms in which disease-associated variants or altered gene expression can be studied under controlled conditions. In many models, mutations affecting pathways such as alpha-synuclein handling, mitochondrial function, or lysosomal degradation contribute to neuronal stress and degeneration, especially in dopamine-producing neurons. These systems help researchers connect genotype to phenotype, test disease mechanisms, evaluate candidate treatments, and compare how inherited or acquired genetic changes influence disease risk and progression.

Murine Model for Parkinson's Disease: from 6-OH Dopamine Lesion to Behavioral Test

Parkinson disease is caused by loss of dopaminergic innervation to the striatum, which can be experimentally induced by 6-OH-dopamine. We describe how to perform a stereotaxic lesion and to monitor apomorphine-induced rotational behavior in mice. This model is useful and reliable for testing new therapies for Parkinson disease.

Histological Examination of Mitochondrial Morphology in a Parkinson's Disease Model

This study presents a method to analyze the morphology of mitochondria based on immunostaining and image analysis in mouse brain tissue in situ. It also describes how this allows one to detect changes in mitochondrial morphology induced by protein aggregation in Parkinson's disease models.

In Parkinson's disease and movement disorders in general, sensitive and reliable behavioral assays are essential for testing novel potential therapeutics. Here, we describe a manageable battery of sensorimotor tests for mice that are sensitive to varying degrees of injury to the nigrostriatal system and useful for preclinical studies.
