Autoantibody Detection

Autoantibody detection is the laboratory identification and measurement of antibodies that mistakenly recognize the body's own cells, tissues, or molecular components, supporting investigation of immune-mediated disease. In common assays, patient serum is exposed to a defined self-antigen or tissue substrate; any binding is then visualized or quantified using enzyme-linked, fluorescent, or other labeled detection reagents. Methods such as ELISA, indirect immunofluorescence, and immunoblotting can reveal antibody specificity, concentration, and cellular staining patterns. In immunology and infection research, these results help characterize autoimmune syndromes, distinguish immune responses associated with infection, assess disease activity, and guide diagnosis or monitoring.

Detection of Human Islet Autoantibodies Using an Electrochemiluminescence Assay

2025

This video showcases the electrochemiluminescence detection of islet autoantibodies in human serum. The antibodies in the serum bridge the ruthenium-conjugated antigen to the biotinylated capture antigen, forming complexes that are detected by electrochemiluminescence measurements.

An Assay to Detect Autoantibodies in Human Serum using a Hippocampal Neuronal Culture

2025

This video demonstrates a technique for identifying antibodies against specific antigens in a sample using a rat hippocampal neuronal culture. By exposing cultured neurons to patient samples containing autoantibodies targeting N-Methyl-D-aspartate receptors (NMDARs), the presence of autoantibodies bound to the neuronal surface is revealed through immunostaining.

Detection of Anti-MDA5 Autoantibodies Using HeLa Cells and Immunocytochemistry with Light Microscopy

2025

We provide a step-by-step immunocytochemistry (ICC) methodology for detecting anti-MDA5. This approach involves cell fixation, permeabilization, antibody incubation, and imaging techniques, which allow for the accurate detection of anti-MDA5 autoantibodies, aiding in the diagnosis of rapidly progressing interstitial lung disease in myositis patients.

Generation of Two-color Antigen Microarrays for the Simultaneous Detection of IgG and IgM Autoantibodies

We describe here a method to generate customizable antigen microarrays that can be used for the simultaneous detection of serum IgG and IgM autoantibodies from humans and mice. These arrays allow for high-throughput and quantitative detection of antibodies against any antigens or epitopes of interest.
