Cell Interaction Analysis

Cell interaction analysis is the study of how cells communicate, recognize one another, and alter their behavior through physical contact or soluble signals. In immunology and infection research, it examines processes such as receptor–ligand binding, cell adhesion, cytokine exchange, and pathogen-mediated changes in signaling using approaches including co-culture, microscopy, and flow cytometry. These measurements can reveal how immune cells coordinate activation, migration, killing, or tolerance and how infectious agents evade or disrupt host defenses. The resulting insights support studies of immune regulation, disease mechanisms, vaccine responses, and the development of targeted antimicrobial or immunomodulatory therapies.

Coculture Analysis of Extracellular Protein Interactions Affecting Insulin Secretion by Pancreatic Beta Cells

Transcellular protein interactions are important determinants of pancreatic beta-cell function. Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

Correlative Microscopy for 3D Structural Analysis of Dynamic Interactions

We describe a correlative microscopy method that combines high-speed 3D live-cell fluorescent light microscopy and high-resolution cryo-electron tomography. We demonstrate the capability of the correlative method by imaging dynamic, small HIV-1 particles interacting with host HeLa cells.

Adhesion Frequency Assay for In Situ Kinetics Analysis of Cross-Junctional Molecular Interactions at the Cell-Cell Interface

An adhesion frequency assay for measuring receptor-ligand interaction kinetics when both molecules are anchored on the surfaces of the interacting cells is described. This mechanically-based assay is exemplified using a micropipette-pressurized human red blood cell as adhesion sensor and integrin αLβ2 and intercellular adhesion molecule-1 as interacting receptors and ligands.

This article describes an experimental approach for dynamic regulation of cell-cell interactions between adherent cells on a micrometer scale. Manipulation of intercellular communication between hepatocytes and stromal cell is demonstrated. The developed platform enables investigation of cell-cell interactions in a variety of biological processes, including development and pathogenesis.
