Epidermal Sheets

Epidermal sheets are thin, multilayered epithelial tissues composed primarily of keratinocytes, produced naturally or engineered in culture to model or restore the skin barrier. During formation, keratinocytes proliferate on a supporting matrix and differentiate into progressively specialized layers, often after exposure to an air–liquid interface; the resulting tissue limits microbial entry while releasing cytokines and antimicrobial factors. In immunology and infection research, epidermal sheets provide controlled systems for studying pathogen invasion, barrier disruption, inflammatory signaling, and host defense, while clinical grafts can support wound coverage and tissue repair.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.
