Folate Receptor Alpha-1

Folate receptor alpha-1 (FOLR1) is a high-affinity cell-surface protein that binds folate and helps cells acquire this vitamin for essential metabolic processes. Anchored to the plasma membrane by a glycosylphosphatidylinositol linkage, it captures folate and delivers it into cells through receptor-mediated endocytosis, supporting one-carbon metabolism, nucleotide production, and methylation reactions. In immunology and infection research, studying FOLR1 helps clarify how folate availability and cellular uptake influence host-cell function, proliferation, and responses to physiological stress. Its selective expression and internalization pathway also make it relevant to biomarker development and targeted delivery of therapeutic or imaging agents.

Ovarian cancer forms metastases throughout the peritoneal cavity. Here, we present a protocol to make and use folate-receptor targeted surface-enhanced resonance Raman scattering nanoprobes that reveal these lesions with high specificity via ratiometric imaging. The nanoprobes are administered intraperitoneally to living mice, and the derived images correlate well with histology.

The aim of this method is to determine the CR1 density in the erythrocytes of any subject by comparing with three subjects whose erythrocyte CR1 density is known. The method uses flow cytometry after immunostaining of the subjects' erythrocytes by an anti-CR1 monoclonal antibody coupled to an amplified system using phycoerythrin (PE).

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.
