Intracellular Localization

Intracellular localization is the determination of where a molecule, pathogen, or host factor resides within a cell, a key question because location often governs function, access, and immune recognition. Researchers map localization by tagging targets with fluorescent probes or antibodies and imaging cells, while fractionation and colocalization with organelle markers can distinguish compartments such as the cytosol, nucleus, endosomes, and lysosomes. In immunology and infection, these approaches reveal how microbes enter, replicate, evade detection, or encounter antimicrobial pathways, and how immune proteins move during activation. Such information supports mechanism-based studies of pathogenesis, host defense, and therapeutic targeting.

Localization of Plasma Membrane and Intracellular Neuronal Nicotinic Acetylcholine Receptors Using Quantitative Imaging in Mammalian Cells

This protocol describes expression and quantification of neuronal nicotinic acetylcholine receptors subunits using the mammalian Neuro-2a cell line. pH-sensitive fluorescent tags and confocal microscopy enable precise evaluation of receptor localization at the plasma membrane versus those expressed in pH-neutral intracellular compartments, facilitating the study of nAChR trafficking and pharmacological modulation.

This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity.
