Intradermal Infection

Intradermal infection is the establishment of a microbial infection within the dermis, the skin layer beneath the epidermis, and is important for understanding how tissue barriers and local immunity respond to invading pathogens. Infection may arise when microorganisms are introduced through a skin breach or inoculation, triggering recognition by resident immune cells and recruitment of inflammatory cells, followed by activation of adaptive immune responses. In immunology and infection research, intradermal infection models help researchers examine pathogen dissemination, lesion development, host defense, and immune-mediated tissue damage under controlled conditions, supporting evaluation of vaccines, therapeutics, and mechanisms of protective immunity.

Myeloid Cell Isolation from Mouse Skin and Draining Lymph Node Following Intradermal Immunization with Live Attenuated Plasmodium Sporozoites

We describe here a protocol for isolating myeloid cells from mouse skin and draining lymph node following intradermal injection of Plasmodium sporozoites. Flow cytometry of collected cells provides a reliable assay to characterize the skin and draining lymph node inflammatory response to the parasite.
