Ldh Sequestration Analysis

LDH sequestration analysis is a biochemical approach for determining how lactate dehydrogenase (LDH) is retained within cells or distributed among cellular compartments during immune and infectious responses. The method typically compares LDH enzymatic activity in intact cells, separated fractions, and surrounding medium, using the enzyme’s conversion of lactate and NAD+ to pyruvate and NADH as a measurable readout. These measurements can help distinguish altered cellular metabolism from membrane damage and assess how pathogens, immune activation, or cytotoxic treatments affect cell integrity and metabolic organization. The results support host-pathogen studies and improve interpretation of LDH-based cytotoxicity assays.

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
