Lymphocyte Migration

Lymphocyte migration is the directed movement of immune lymphocytes through blood, lymph, and tissues, a process essential for immune surveillance and effective responses to infection. Chemokines create concentration gradients that guide lymphocytes, while selectins initiate rolling along vascular endothelium and integrins strengthen adhesion, allowing cells to cross the vessel wall by diapedesis. Once in lymphoid organs or infected tissues, migration helps T and B cells locate antigen-presenting cells, coordinate inflammation, and eliminate pathogens. Studying this process clarifies how immune defenses are organized and supports research into inflammatory disease, immunodeficiency, cancer, and therapies that alter leukocyte trafficking.

Assessment of Lymphocyte Migration in an Ex Vivo Transmigration System

In this protocol, lymphocytes are placed in the top chamber of a transmigration system, separated from the bottom chamber by a porous membrane. Chemokine is added to the bottom chamber, which induces active migration along a chemokine gradient. After 48 h, lymphocytes are counted in both chambers to quantitate transmigration.

The video demonstrates an immunological method to monitor in vivo adjuvant-induced cytotoxic T lymphocyte or CTL production. A mouse is pre-injected with genetically engineered, proliferative dye-stained donor CD8+ T lymphocytes. Subcutaneous injection of ovalbumin with adjuvants generates cytotoxic CD8+ T lymphocytes that are identified post-harvesting through immunostaining and flow cytometry.

Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.
