Mosquito Cell Transfection

Mosquito cell transfection is a laboratory technique used to introduce foreign nucleic acids into mosquito-derived cells, enabling researchers to study gene function, vector biology, and pathogen interactions. During transfection, DNA or RNA enters cultured cells through a delivery method such as lipid-based carriers or electroporation, allowing the introduced genetic material to be expressed or used to alter cellular activity. In immunology and infection research, the technique supports analysis of antiviral responses, mosquito immune pathways, vector competence, and host-pathogen interactions. It can also facilitate production of recombinant proteins and evaluation of genetic targets that may influence disease transmission.

We report using an artificial intronic small RNA expression strategy to develop a non-defective recombinant Aedes aegypti densovirus (AaeDV) in vivo delivery system. A detailed procedure for the construction, packaging, and quantitative analysis of the rAaeDV vectors as well as for larval infection is described.
