Pbmc Cryopreservation

PBMC cryopreservation is the controlled storage of peripheral blood mononuclear cells, primarily lymphocytes and monocytes, at very low temperatures so samples remain available for later immunological analysis. Cells are suspended in a cryoprotective medium, commonly containing dimethyl sulfoxide (DMSO), cooled at a controlled rate to limit intracellular ice formation, and stored in liquid nitrogen or its vapor phase. During recovery, rapid thawing, dilution or washing, and careful handling help reduce osmotic and cryoprotectant-related injury while preserving cell viability and function. In immunology and infection research, preserved PBMCs support standardized immune assays, pathogen-response studies, biobanking, and comparisons across time points or cohorts.

Mouse Sperm Cryopreservation and Recovery using the I·Cryo Kit

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

A Technique to Generate a Human PBMC-Engrafted Humanized Xenograft Mouse Model

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

Cryopreservation of Preimplantation Embryos of Cattle, Sheep, and Goats

Preimplantation embryos may be cryopreserved after placement into a hypertonic cryoprotective solution to cause cellular dehydration. After equilibration, ice crystal formation is induced in the solution surrounding the embryo. Further dehydration occurs as the embryo is slowly cooled to subzero temperatures before plunging into liquid nitrogen for storage.
