Phagocytic Index Calculation

Phagocytic Index Calculation is a quantitative method for assessing how effectively phagocytes ingest microbes or other particulate targets, making it useful for evaluating innate immune function. The index is typically derived by examining stained phagocytes under a microscope and relating the number of engulfed particles to the number of cells analyzed, often distinguishing internalized material from particles merely attached to the cell surface. In immunology and infection research, this measurement helps compare phagocytic activity across cell types, treatments, pathogens, or patient samples. It can reveal changes in host defense, support antimicrobial studies, and provide a standardized outcome for investigating immune dysfunction.

We present a high-throughput flow cytometric assay to determine the phagocytic activity of antigen-specific antibodies from clinical samples, utilizing fluorescent antigen-coated beads and a monocytic cell line expressing multiple Fc receptors—providing receptor usage and phagocytic activity determinations in a standardized and reproducible fashion for any antigen of interest.
