Pneumococcal Isolate

A pneumococcal isolate is a laboratory-recovered strain of Streptococcus pneumoniae, the encapsulated bacterium responsible for pneumonia, meningitis, sepsis, and other infections. Laboratories obtain isolates from clinical specimens and identify them through culture characteristics, including alpha-hemolytic growth, together with tests such as optochin susceptibility, bile solubility, and molecular or biochemical analysis. Characterizing a pneumococcal isolate helps determine serotype, antimicrobial susceptibility, and virulence-related features, supporting diagnosis, epidemiological surveillance, vaccine evaluation, and studies of host-pathogen interactions. In immunology and infection research, isolates provide experimentally defined material for examining capsule-mediated immune evasion and responses to pneumococcal infection.

Streptococcus pneumoniae is the leading pathogen causing severe community-acquired pneumonia and responsible for over 2 million deaths worldwide. The impact of bacterial factors implicated in fitness or virulence can be monitored in real-time in an acute mouse pneumonia or bacteremia model using bioluminescent bacteria.

Source: Jagau, H. et al. Pneumococcus Infection of Primary Human Endothelial Cells in Constant Flow. J. Vis. Exp. (2019)This video demonstrates the real-time visualization of pneumococcal attachment to Von Willebrand Factor (VWF) strings on endothelial cells under constant media flow conditions. It outlines the steps for stimulating VWF release, labeling the strings, and quantifying bacterial binding.

Using the Overlay Assay to Qualitatively Measure Bacterial Production of and Sensitivity to Pneumococcal Bacteriocins

Competition in Streptococcus pneumoniae is mediated by bacteriocins, small antimicrobial peptides with inhibitory activity towards pneumococcus and other related species. Here we describe an optimized bacterial overlay assay that allows for the characterization of bacteriocin activity and inhibitory spectrum, bacteriocin-specific immunity, and detection of secreted quorum sensing peptides.
