Serial Two-photon Tomography

Serial two-photon tomography is a three-dimensional imaging technique that combines two-photon fluorescence microscopy with serial physical sectioning to map biological structures through large tissue volumes. A focused infrared laser excites fluorescent labels near the exposed tissue surface, while automated sectioning removes thin layers and enables repeated imaging for volumetric reconstruction with reduced out-of-focus fluorescence. In immunology and infection research, the method can reveal the spatial organization of immune cells, pathogens, and tissue microenvironments across intact organs. These maps help connect cellular interactions and infection-associated remodeling to tissue-scale outcomes, complementing molecular analyses and supporting more complete models of host-pathogen responses.

Diffuse fluorescence tomography offers a relatively low-cost and potentially high-throughout approach to preclinical in vivo tumor imaging. The methodology of optical data collection, calibration, and image reconstruction is presented for a computed tomography-guided non-contact time-domain system using fluorescent targeting of the tumor biomarker epidermal growth factor receptor in a mouse glioma model.
