Collagen Embedding

Collagen embedding is a technique that suspends cells, tissues, or biological specimens within a collagen-based matrix to create a supportive three-dimensional environment for medical research. The method typically uses soluble collagen that polymerizes into a fibrillar hydrogel under controlled pH and temperature, forming a scaffold that permits cell attachment, migration, and interaction with the surrounding matrix. This approach helps model tissue structure and behavior more realistically than flat culture surfaces and supports studies of wound repair, tissue engineering, disease progression, and therapeutic responses. Collagen embedding therefore connects basic cell biology with the development and evaluation of regenerative and diagnostic strategies.

This manuscript describes a soft lithography-based technique to engineer uniform arrays of three-dimensional (3D) epithelial tissues of defined geometry surrounded by extracellular matrix. This method is amenable to a wide variety of cell types and experimental contexts and allows for high-throughput screening of identical replicates.
