Collagen Extraction

Collagen extraction is the process of isolating collagen from animal or tissue sources for study and medical use. It typically involves pretreatment to remove fats and noncollagenous proteins, followed by acidic or enzymatic solubilization, often with pepsin, which cleaves terminal regions and releases collagen while preserving much of its triple-helical structure. The extracted material can then be purified, characterized, and formed into films, gels, sponges, or scaffolds. In medicine, these collagen-based materials support research and development in wound dressings, tissue engineering, drug delivery, and regenerative therapies, where biocompatibility and structural properties are important.

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.

Imaging Denatured Collagen Strands In vivo and Ex vivo via Photo-triggered Hybridization of Caged Collagen Mimetic Peptides

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.
