Cytokine Multiplex Assay

Cytokine multiplex assays are laboratory methods that measure multiple cytokines simultaneously in a single biological sample, providing a broader view of immune activity than single-analyte tests. In a typical bead-based format, distinct bead populations coated with capture antibodies bind specific cytokines, while detection antibodies and fluorescent signals identify and quantify each analyte. In medicine, these assays support evaluation of inflammation, infection, immune disorders, cancer, and treatment responses using samples such as blood or other body fluids. By revealing coordinated cytokine patterns, they can help characterize disease mechanisms, identify biomarkers, and guide research into personalized therapies.

We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.
