Dissecting Microscope Analysis

Dissecting microscope analysis is the examination of relatively large, three-dimensional specimens with a stereomicroscope, allowing researchers and clinicians to assess surface features, structure, and spatial relationships at low magnification. The instrument uses separate optical paths for the viewer’s eyes to create depth perception, while reflected illumination reveals external details without requiring thin tissue sections. In medicine, this approach supports gross dissection, anatomical inspection, pathology specimen preparation, and the handling of tissues or small biological structures during laboratory procedures. Its combination of three-dimensional viewing, working distance, and modest magnification helps connect macroscopic observations with findings from histology and other diagnostic methods.

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.
