Glioblastoma Tumor Microenvironment

The glioblastoma tumor microenvironment is the network of noncancerous cells, extracellular matrix, blood vessels, and soluble signals surrounding and interacting with glioblastoma cells. Hypoxia and tumor-derived signals can recruit and reprogram microglia and macrophages, alter astrocyte and vascular cell behavior, and promote cytokine release, matrix remodeling, invasion, immune suppression, and resistance to therapy. Characterizing these interactions helps researchers understand why glioblastoma progresses rapidly and responds poorly to treatment. Tumor microenvironment models also support the identification of biomarkers and therapeutic strategies aimed at disrupting angiogenesis, immune suppression, or stromal support, potentially improving drug development and patient outcomes.

We present the procedure for fabrication and operation of a microfluidic device that recreates heterogeneous tumor microenvironments in vitro. The variability in apoptosis within tumor tissue was quantified using fluorescent stains and the effective diffusion coefficient of the chemotherapeutic drug doxorubicin into tumor tissue was evaluated.

Tumor Treating Field Therapy in Combination with Bevacizumab for the Treatment of Recurrent Glioblastoma

A novel methodology that is employed for the treatment of recurrent glioblastomas is described. This treatment approach employs the application of alternating electric tumor treating fields (TTFields), known as TTF Therapy in combination with bevacizumab, a targeted agent that is currently FDA approved as monotherapy.
