In Vitro Eye Models

In vitro eye models are laboratory-based systems that reproduce selected structures or functions of the eye outside a living organism, providing controlled platforms for studying ocular biology and disease. Researchers create them from cultured cells, tissue explants, or stem-cell-derived organoids and regulate variables such as cell type, extracellular environment, and exposure to drugs or injury; measured changes in cell behavior, barrier function, or tissue morphology reveal underlying mechanisms. These models support investigation of retinal, corneal, and other eye disorders, evaluation of therapeutic candidates, and assessment of toxicity before clinical studies. Their controlled design can complement animal research and improve the efficiency and relevance of biomedical testing.

Eye Irritation Test (EIT) for Hazard Identification of Eye Irritating Chemicals using Reconstructed Human Cornea-like Epithelial (RhCE) Tissue Model

We have developed an eye irritation test which utilizes a three dimensional reconstructed human cornea-like epithelial (RhCE) tissue model. The test is able to discriminate between ocular irritant and corrosive materials (GHS Categories 1 and 2 combined) and those that do not require labeling (GHS No Category).

This procedure was established to be used for developing advanced 3D hepatic cultures in vitro, which can provide a more physiologically relevant assessment of the genotoxic hazards associated with nanomaterial exposures over both an acute or long-term, repeated dose regimes.

This report describes a method to induce chronic experimental autoimmune dry eye in Lewis rats through immunization with an emulsion of rat lacrimal gland extract, ovalbumin, and complete Freund's adjuvant, followed by the injection of lacrimal gland extract and ovalbumin into the forniceal subconjunctiva and lacrimal glands six weeks later.

A primary culture of bovine corneal endothelial cells was used to investigate the mechanism of corneal endothelial-mesenchymal transition. Furthermore, a rat corneal endothelium cryoinjury model was used to demonstrate corneal endothelial-mesenchymal transition in vivo.
