Mcf10a Cells

MCF10A cells are a spontaneously immortalized, non-tumorigenic human mammary epithelial cell line widely used to model normal breast-cell biology and early changes associated with cancer. In culture, their proliferation depends on defined growth conditions containing factors such as epidermal growth factor and insulin, while three-dimensional extracellular-matrix systems can induce organized acinar structures and growth arrest under appropriate conditions. These characteristics make MCF10A cells useful for studying epithelial morphogenesis, cell polarity, adhesion, migration, and signaling responses in medicine. Researchers also use them as a nonmalignant comparison model when evaluating oncogenic transformation, drug effects, and mechanisms of breast cancer progression.

Here, we describe xenograft zebrafish models using two different injection sites, i.e., perivitelline space and duct of Cuvier, to investigate the invasive behavior and to assess the intravasation and extravasation potential of human breast cancer cells, respectively.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.
