Patterned Cell Culture

Patterned cell culture is a technique that positions cells in defined spatial arrangements on laboratory substrates, enabling researchers to study how organization influences cell behavior and tissue function. It typically uses micropatterned surfaces, selective cell-adhesion regions, or physical barriers to control where cells attach, spread, and interact under controlled culture conditions. In medicine, patterned cell culture helps model tissue architecture, investigate cell-cell and cell-matrix interactions, and examine processes such as differentiation, migration, and tissue formation. These controlled systems can improve disease modeling, drug testing, and the development of engineered tissues by producing more physiologically relevant cellular environments.

Aqueous two-phase systems were used to simultaneously pattern multiple populations of cells. This fast and easy method for cell patterning takes advantage of the phase separation of aqueous solutions of dextran and polyethylene glycol and the interfacial tension that exists between the two polymer solutions.

We describe a sequential process for light-mediated formation and subsequent biochemical patterning of synthetic hydrogel matrices for three-dimensional cell culture applications. The construction and modification of hydrogels with cytocompatible photoclick chemistry is demonstrated. Additionally, facile techniques to quantify and observe patterns and determine cell viability within these hydrogels are presented.
