Pharmacologic Melanin Induction

Pharmacologic melanin induction is the use of drugs or drug-like compounds to increase melanin production in melanocytes, the pigment-forming cells that influence skin, hair, and eye color. These agents act through melanogenic signaling pathways, often enhancing melanocortin receptor activity or increasing the expression and activity of tyrosinase, the rate-limiting enzyme in melanin synthesis; newly formed pigment can then be packaged into melanosomes and transferred to keratinocytes. In medicine, this approach is studied for pigmentary disorders such as vitiligo, controlled photoprotection, and research into melanocyte biology. Clinical use requires attention to treatment response, durability, and potential adverse effects.

Epidermal melanin is induced by topical application of forskolin in a murine model of the fair-skinned UV-sensitive human. Pharmacologic manipulation of cAMP levels in the skin and epidermal darkening strongly protect against UV-mediated inflammation (sunburn) as measured by the minimum erythematous dose (MED) assay.

Loss of the righting reflex has long served as a standard behavioral surrogate for unconsciousness, also called hypnosis, in laboratory animals. Alterations in volatile anesthetic sensitivity caused by pharmacological interventions can be detected with a carefully controlled high-throughput assessment system, which may be adapted for delivery of any inhaled therapeutic.
