Tear Lipidomics

Tear lipidomics is the comprehensive analysis of lipid molecules in the tear film, providing a molecular view of the ocular surface and its protective functions. In medicine, researchers collect tear samples, extract their lipid content, and use chromatographic separation with mass spectrometry to identify and quantify lipid classes such as wax esters, cholesteryl esters, phospholipids, and sphingolipids. These measurements help characterize the tear-film lipid layer, which limits evaporation and supports surface stability. Comparing lipid profiles can clarify mechanisms of dry eye disease, inflammation, and meibomian gland dysfunction, while supporting biomarker discovery and evaluation of targeted treatments.

2025

This protocol describes a unique clinical protocol for collecting human tear fluid samples using phenol red threads and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based workflow to discover the tear lipidomic profile. The simple methyl tert-butyl ether (MTBE)/methanol biphasic separation method enables rapid tear lipid extraction with high recovery for tear biomarker discovery.

Capsulorhexis is an important step in phacoemulsification surgery. A surgeon creates a continous curvilinear tear on the anterior lens capsule by controlling the tearing vector forces. A peripherally extended tear is a serious complication. This video demonstrates a novel technique of rescuing capsular radial tear out using a cystotome.
