Demyelinating Disease Model

A demyelinating disease model is an experimental system that reproduces the loss or damage of myelin, the insulating layer that enables efficient communication between nerve cells. In neuroscience research, models can induce demyelination through immune-mediated injury, toxic compounds, genetic changes, or targeted damage to oligodendrocytes, allowing researchers to examine myelin loss and subsequent remyelination. These models help clarify how disrupted axonal conduction contributes to neurological dysfunction and how inflammatory responses affect repair. They also support evaluation of candidate therapies, including treatments designed to protect oligodendrocytes, reduce inflammation, restore myelin, and improve functional recovery in disorders such as multiple sclerosis.

Establishing a Focal Demyelination Mouse Model via a Two-Point Injection of Lysophosphatidylcholine

2025

This video demonstrates the generation of a focal demyelination mouse model using a two-point Lysophosphatidylcholine (LPC) injection. The skull of an anesthetized mouse is exposed. The injection coordinates are determined, and a hole is drilled to enable LPC injections at targeted sites. LPC induces focal demyelination of neurons via inflammation, while a two-point injection ensures its sustainability.

(2021)This video demonstrates the method for delivering neurotherapeutic drugs using an osmotic pump in a demyelinated mouse model. The controlled drug release promotes remyelination by activating myelin-producing cells in the corpus callosum.

Generating the Widespread Cerebral Cortical Demyelination in a Rat Model

2025

The video demonstrates a method for generating a rat model with widespread cerebral cortical demyelination. This involves disrupting the blood-brain barrier by injecting pro-inflammatory cytokines into mice primed with recombinant myelin sheath protein. This process facilitates the infiltration of immune cells, resulting in widespread cerebral cortical demyelination.

Focal demyelination is induced in the optic nerve using lysolecithin microinjection. Visual evoked potentials are recorded via skull electrodes implanted over the visual cortex to examine the signal conduction along the visual pathway in vivo. This protocol details the surgical procedures underlying electrode implantation and optic nerve microinjection.
