Methyl Salicylate

Methyl salicylate is an aromatic ester found in wintergreen oil that serves as a flavoring compound, topical analgesic, and laboratory reagent. In neuroscience, its solvent properties and relatively high refractive index allow dehydrated, fixed tissue to become optically transparent by reducing light scattering, enabling microscopic visualization of neural structures. Researchers have used methyl salicylate-based clearing approaches to examine brain and nervous-system anatomy, trace neuronal morphology, and document connections in three dimensions. Although newer aqueous and solvent-based methods offer improved compatibility with fluorescent labels, methyl salicylate remains a useful, accessible clearing medium for selected anatomical studies and historical microscopy workflows.

DNA Methylation: Bisulphite Modification and Analysis

The gold standard for DNA methylation analysis is genomic sequencing of bisulphite converted DNA. This method takes advantage of the increased sensitivity of cytosine compared with 5-methylcytosine (5-MeC) to bisulphite deamination under acidic conditions. Unmethylated cytosines can be distinguished from methylated cytosines after PCR amplification of the target genomic DNA.
