Olfactory Sensillum Analysis

Olfactory sensillum analysis is the examination of specialized sensory structures that detect odorants and encode chemical information in the nervous system. It typically combines anatomical characterization with recordings from individual sensilla: odor molecules enter surface pores, interact with olfactory receptor neurons, and alter their electrical activity, producing spike patterns measured during controlled odor stimulation. In neuroscience, this approach links sensillum structure and receptor function to odor coding, helping researchers compare sensitivity, response dynamics, and selectivity across sensory neurons. These measurements support studies of chemosensation, neural representation, and how animals distinguish biologically relevant smells.

Electrophysiological responses of olfactory sensory neurons to odorants can be measured in insects using single sensillum recordings. In this video article we will demonstrate how to perform single sensillum recordings in the antennae of the vinegar fly (Drosophila melanogaster) and the maxillary palps of the malaria mosquito (Anopheles gambiae).

High-throughput Analysis of Mammalian Olfactory Receptors: Measurement of Receptor Activation via Luciferase Activity

Olfactory receptor activation patterns encode odor identity, but the lack of published data identifying odorant ligands for mammalian olfactory receptors hinders the development of a comprehensive model of odor coding. This protocol describes a method to facilitate high-throughput identification of olfactory receptor ligands and quantification of receptor activation.

Fundamental, yet unique properties of the rodent olfactory system have led to its increasing study among biologists. A relatively simple assessment of its function is then also needed. Here we describe sensitive tests for the characterization of mouse olfactory sensitivity and preference.

Quadruple Immunostaining of the Olfactory Bulb for Visualization of Olfactory Sensory Axon Molecular Identity Codes
