Peptide Identification

Peptide identification is the process of determining the amino acid sequence and molecular identity of peptides in a biological sample, a key step in linking neurochemical signals to neuronal function. It commonly uses liquid chromatography coupled with tandem mass spectrometry, where peptides are separated, ionized, fragmented into sequence-informative product ions, and matched with theoretical or reference spectra. In neuroscience, this approach helps identify neuropeptides and peptide hormones in brain tissue, cerebrospinal fluid, or neuronal cultures, supporting studies of synaptic communication, stress, pain, appetite, and disease-related signaling. Reliable identification can guide functional experiments and biomarker research.

Techniques to dissect the mechanisms underlying the secretion of HIV-1 Nef in exosomes are described. Specific short peptides derived from Nef and protein transfection were exploited to determine the structure, function, and binding partners of Nef’s Secretion Modification Region. These procedures have general relevance in many mechanistic studies.
