Postnatal Gene Delivery

Postnatal gene delivery is the introduction of therapeutic or experimental genetic material into an organism after birth, enabling researchers to modify gene expression in the developing or mature nervous system. In neuroscience, delivery commonly uses viral vectors such as adeno-associated viruses or nonviral systems administered by local injection or other routes; the payload enters target cells, where vector elements support transient or sustained expression while tissue access and immune responses influence efficiency. This approach supports studies of neuronal function, circuit development, and disease mechanisms, and may enable treatments for inherited neurological disorders by restoring, silencing, or regulating specific genes.

An in vivo method to test gene function in postnatal brain is described. Recombinant AAVs expressing Cre and/or a fluorescent protein are injected into neonatal mouse brain. Mosaic gene inactivation and sparse neuronal labeling are achieved, allowing rapid analysis of gene function in processes critical to neural circuit development.
