Retinal Vascular Quantification

Retinal vascular quantification is the measurement and analysis of blood-vessel structure and distribution in the retina, providing objective indicators of vascular and neural health. It typically uses retinal photographs or optical coherence tomography angiography images, followed by image segmentation and computational analysis of features such as vessel density, caliber, branching, and tortuosity. In neuroscience, these measurements help characterize neurovascular coupling and detect vascular alterations associated with neurological and ophthalmic disease. Because retinal vessels can be assessed noninvasively, quantitative vascular metrics support disease monitoring, evaluation of treatment effects, and research into links between retinal circulation and central nervous system function.

This protocol describes a method to study dementia using retinal imaging techniques. This method can potentially aid in diagnosis and risk assessment of dementia.

The pericytes in retinal vasculature were examined by immunofluorescent staining with platelet-derived growth factor receptor β after retro-orbital injection of fluorescent tomato lectin. The labeled retina was further treated with the tissue-clearing method and whole mounted for visualizing the three-dimensional views of pericytes surrounding retinal vasculature under a confocal microscope.
