Tissue Preparation Method

Tissue preparation methods are procedures that preserve, process, and arrange biological tissue for microscopic or molecular analysis, making cellular structures and disease-related changes measurable. In neuroscience, samples commonly undergo fixation to stabilize proteins and membranes, followed by dehydration, embedding or cryoprotection, sectioning, and staining; each step helps maintain tissue architecture and expose specific cellular features. Proper preparation supports visualization of neurons, glial cells, synapses, axonal pathways, and pathological lesions using light or fluorescence microscopy. Consistent protocols improve reproducibility across brain research, enabling studies of development, connectivity, neurodegeneration, and responses to injury or treatment.

Corneal Donor Tissue Preparation for Endothelial Keratoplasty

Endothelial corneal transplantation is a surgical technique for treatment of posterior corneal diseases. Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes. Dissections can be performed at the eye bank prior to corneal transplantation surgery.

We describe a protocol for transcardiac perfusion of mice, removal and sectioning of the brain, as well as immunoperoxidase staining, resin embedding, and ultrathin sectioning of the brain sections. Upon completion of these procedures, the immunostained material is ready for examination with transmission electron microscopy.

Nanoscaled sea-island surfaces composed of thermoresponsive block copolymers were fabricated by the Langmuir-Schaefer method for controlling spontaneous cell adhesion and detachment. Both the preparation of the surface and the adhesion and detachment of cells on the surface were visualized.
