Trans Adhesion Assay

A trans adhesion assay is a cell-based or protein-based method for measuring whether adhesion molecules bind across opposing surfaces, a key process in establishing and maintaining contacts between neurons. In the assay, one adhesion partner is presented on one cell or substrate and a potential binding partner on another; their interaction brings the surfaces together, allowing adhesion strength or binding specificity to be quantified. In neuroscience, this approach helps characterize trans-synaptic proteins, examine how molecular recognition organizes pre- and postsynaptic sites, and compare disease-associated or engineered variants. The results clarify mechanisms of synapse formation, connectivity, and neural circuit development.

Cell Aggregation Assay to Detect Trans Interactions between Cell Adhesion Molecules

2025

In this video, we demonstrate the technique to measure transcellular interactions between synaptic cell adhesion molecules through the HEK cell aggregation assay. In this assay, an equal number of HEK cells expressing a desired presynaptic cell adhesion molecule and a fluorescent protein are mixed with HEK cells expressing a post-synaptic cell adhesion molecule and another fluorescent protein. The interaction between the cell adhesion molecules forces the cells to aggregate, which can be...

Assay of Adhesion Under Shear Stress for the Study of T Lymphocyte-Adhesion Molecule Interactions

This flow adhesion assay provides a simple, high impact model of T cell-epithelial cell interactions. A syringe pump is used to generate shear stress, and confocal microscopy captures images for quantification. The goal of these studies is to effectively quantify T cell adhesion using flow conditions.

We describe a valuable diagnostic assay that could potentially be used to decide the withdrawal of immunosuppression after transplant without elevated risk of graft rejection. The assay uses the principles of Delayed Type Hypersensitivity and provides accurate assessment of both donor specific effector and regulatory immune responses mounted by recipients.

A method of quantitating neutrophil adhesion is reported. This method creates a dynamic flow environment similar to that encountered in a blood vessel. It allows the investigation of neutrophil adhesion to either purified adhesion molecules (ligand) or endothelial cell substrate (HUVEC) in a context similar to the in vivo environment with sheer stress.

We describe a qualitative assay for yeast adhesion and agar invasion as a measure of invasive and pseudohyphal differentiation. This simple assay can be used to assess the invasive phenotype of various mutants as well as the effects environmental cues and signaling pathways on yeast differentiation.
