Benign Prostatic Hyperplasia

Benign prostatic hyperplasia (BPH) is a noncancerous enlargement of the prostate that can obstruct urinary flow and cause lower urinary tract symptoms, including hesitancy, weak stream, and increased frequency. Symptoms arise from both increased prostate tissue, driven partly by dihydrotestosterone, and heightened smooth-muscle tone at the prostate and bladder neck. Pharmacological treatment targets these mechanisms: alpha-1 adrenergic antagonists relax smooth muscle for relatively rapid relief, while 5-alpha-reductase inhibitors reduce dihydrotestosterone formation and gradually shrink the prostate. Understanding these therapies helps guide symptom management, prevent complications such as urinary retention, and support individualized treatment decisions.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.
