Conditioned Place Preference

Conditioned place preference is a behavioral assay that measures whether an animal develops an attraction or aversion to an environment associated with a specific stimulus. During conditioning, the animal repeatedly receives a drug or other treatment in one chamber and a control treatment in a distinct chamber; a later drug-free preference test reveals whether contextual cues have acquired motivational value through associative learning. In pharmacology, this method helps evaluate rewarding and aversive drug effects, investigate neural mechanisms underlying reinforcement and addiction, and assess how experimental compounds influence drug-associated behavior. Results can guide studies of dependence, relapse, and potential therapeutic interventions.

This protocol describes the Conditioned Place Preference (CPP) as a model of relapse. This procedure permits the measurement of relapse in laboratory animals, considering the impact of drug-associated environmental cues as craving and relapse in abstaining addicts is currently the focus of drug-abuse treatment programs.

We describe a technique to evaluate song preference in zebra finches. Females are placed in a two-chambered cage and song preference is measured by the number of times she triggers the playback of one song by landing on a perch within one chamber, compared with triggering a different song in the second chamber. Perch landings are counted using infrared sensors.
