In Situ Analysis

In situ analysis is the examination of a material, system, or physical process in its original location or under the conditions in which it operates, preserving context that laboratory removal may alter. In physics, researchers combine real-time measurements with controlled variables such as temperature, pressure, electric or magnetic fields, while probes, detectors, imaging, or spectroscopy record changes as they occur. This approach links structure and behavior, helping identify phase transitions, mechanical responses, transport phenomena, and dynamic interactions. In situ analysis supports research in condensed matter, materials science, nanotechnology, and device development by revealing mechanisms that static or post-process measurements can miss.

We describe a procedure for the detection of chemical elements present in situ in human cells as well as their in vitro quantification. The method is well-suited to any cell type and is particularly useful for quantitative chemical analyses in single cells following in vitro metal oxide nanoparticles exposure.

We present a protocol for structural and compositional analysis of natural oral biofilm from orthodontic appliances with in situ hybridization (FISH) and confocal laser scanning microscopy (CLSM). Oral biofilm samples were collected from palatal expanders, scraping acrylic-resin flakes off their surface and referring them for molecular processing.

Analyses of the sulfur isotopic composition (δ34S) of pyrite from methane-bearing sediments have typically focused on bulk samples. Here, we applied secondary ion mass spectroscopy to analyze the δ34S values of various pyrite generations to understand the diagenetic history of pyritization.
