La transcripción está regulada por activadores y represores, proteínas que se unen al ADN. En los eucariotas, estas proteínas reguladoras a menudo necesitan proteínas adicionales, llamadas correguladores, para funcionar correctamente.
Estos correguladores se unen a complejos activadores o represores; sin embargo, no reconocen secuencias cis-reguladoras, ya que no pueden unirse directamente al ADN. 
Un regulador transcripcional se une a una secuencia cis-reguladora antes de que el co-regulador se asocie; los reguladores y correguladores a menudo no pueden formar complejos estables a menos que el regulador esté unido al ADN. Co-activators and Co-repressors

La transcripción genética está regulada por la acción sinérgica de varias proteínas que forman un complejo en un sitio regulador del gen. Las histonas acetiltransferasas y las histonas desmetilasas funcionan como coactivadores; sin embargo, primero deben localizarse en el sitio regulador mediante un activador de la transcripción para poder realizar estas funciones. Las histonas acetiltransferasas pueden acetilar los residuos de lisina en las colas de histonas. La acetilación desenrolla la cromatina y promueve la expresión genética. Por otro lado, las histonas desacetilasas y las histonas metiltransferasas funcionan como correpresores. Ambas modificaciones conducen a un endurecimiento de la estructura de la cromatina y, por tanto, a la prevención de la expresión genética.

Chromatin control plays a major role in gene transcription in eukaryotes. Gene expression is regulated by several proteins that work together at a gene regulatory site. Some of these proteins bind directly to specific DNA sequences, while co-regulators attach to those regulators but do not bind DNA on their own.

Co-regulators can act as co-activators or co-repressors, depending on the complex they join. One co-regulator may even switch roles in different settings. For example, the transcriptional co-repressor G9a helps activate a steroid hormone receptor gene with co-activators such as GRIP1 and CARM1. Different protein domains help carry out these different jobs.

Many of these regulators also have enzyme activity that changes chromatin structure. Histone acetyltransferases and histone demethylases act as co-activators, but they must first be brought to the regulatory site by a transcription activator. Histone acetyltransferases add acetyl groups to lysine residues on histone tails, which loosens chromatin and supports gene expression.

Other enzymes act as co-repressors. Histone deacetylases and histone methyltransferases tighten chromatin structure and reduce gene expression. These chromatin changes help explain how co-regulators can either turn genes on or keep them off.
