Agar Plate Bioassay

Agar plate bioassay is a microbiological method for measuring the biological activity or concentration of a substance by observing its effect on organisms growing in an agar medium. In a typical assay, an indicator microorganism is embedded in or spread across the agar, while the test sample diffuses from a well or disk and inhibits growth, producing a measurable zone of inhibition. Researchers compare zone diameter with standards to estimate antimicrobial potency or evaluate compounds and biological products. The method supports biochemical quality control and screening, although diffusion, incubation conditions, and organism sensitivity must be standardized for reliable results.

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
