Tagged Protein Expression

Tagged protein expression is a molecular biology technique that produces recombinant proteins fused to short peptide or larger protein tags, enabling their purification, detection, or localization. Researchers insert a tag-encoding sequence into an expression construct so host cells synthesize the target protein and tag as one polypeptide; affinity tags then bind specific ligands during purification, while epitope or fluorescent tags support antibody-based detection or imaging. In biochemistry, this approach streamlines protein isolation and analysis, helping researchers assess protein structure, interactions, activity, and cellular distribution. Careful tag selection and placement can improve experimental yield while minimizing effects on protein folding or function.

SNAP-tag and CLIP-tag protein labeling systems enable the specific, covalent attachment of molecules, including fluorescent dyes, to a protein of interest in live cells. Once cloned and expressed, the tagged protein can be used with a variety of substrates for numerous downstream applications without having to clone again.

Cellular processes such as cell migration have traditionally been studied on two-dimensional, stiff plastic surfaces. This report describes a technique for directly visualizing protein localization and analyzing protein dynamics in cells migrating in a more physiologically relevant, three-dimensional matrix.
