Tyrosine Sulfation

Tyrosine sulfation is a post-translational modification in which a sulfate group is added to tyrosine residues on proteins, altering their molecular interactions and biological activity. In the trans-Golgi network, tyrosylprotein sulfotransferases use 3′-phosphoadenosine 5′-phosphosulfate (PAPS) as the sulfate donor, typically modifying proteins destined for secretion or the cell surface. This modification strengthens specific protein-protein interactions, including binding between chemokines and their receptors, and contributes to processes such as cell signaling, immune-cell trafficking, and coagulation. Studying tyrosine sulfation helps explain how extracellular proteins achieve selective recognition and may support research into therapeutic strategies targeting receptor-ligand interactions.

In this video, we demonstrate a procedure for combined Alcian blue and silver staining of sulfated glycosaminoglycan oligosaccharide fragments. This combined staining technique significantly amplifies the bands of glycosaminoglycan polymers of different lengths in polyacrylamide gels.
