3d Ex Vivo

3D ex vivo models are three-dimensional tissues or organ samples maintained outside the living organism while retaining key aspects of their native architecture and function. In controlled laboratory conditions, cells remain organized within extracellular matrix and preserve cell-cell interactions, allowing researchers to study tissue behavior in a more physiologically relevant setting than many two-dimensional cultures. Bioengineers use these models to evaluate biomaterials, investigate disease processes, test therapeutic strategies, and assess tissue responses before in vivo studies. By linking native tissue complexity with experimental control, 3D ex vivo systems support the development of engineered tissues and more predictive research platforms.

Collagen is a core component of the ECM, and provides essential cues for several cellular processes ranging from migration to differentiation and proliferation. Provided here is a protocol for embedding cells within 3D collagen hydrogels, and a more advanced technique for generating randomized or aligned collagen matrices using PDMS microchannels.

Quantitative 3D oxygen maps of murine tumors were imaged noninvasively using Pulse Electron Paramagnetic Resonance. Ultrasound B-mode and Power Doppler were used for anatomy and vascular structure. Images from both modalities were superimposed enabling multi-parametric tumor analysis.

Measuring 3D In-vivo Shoulder Kinematics using Biplanar Videoradiography

Biplane videoradiography can quantify shoulder kinematics with a high degree of accuracy. The protocol described herein was specifically designed to track the scapula, humerus, and the ribs during planar humeral elevation, and outlines the procedures for data collection, processing, and analysis. Unique considerations for data collection are also described.
