Agar Diffusion Assay

The agar diffusion assay is a laboratory method for evaluating the activity of antimicrobial agents by measuring their ability to inhibit microbial growth in an agar medium. In this test, a microorganism is spread across agar, and a sample is placed in a well or on a paper disk; as the compound diffuses through the gel, it may produce a clear zone of inhibition around the application site. The zone’s diameter provides a comparative measure of antimicrobial effectiveness under defined conditions. In bioengineering, the assay supports screening of antibiotics, antimicrobial biomaterials, coatings, and drug delivery systems, helping assess performance during product development and quality evaluation.
