Endothelial Fibroblast Constructs

Endothelial fibroblast constructs are engineered three-dimensional tissues that combine blood vessel-forming endothelial cells with extracellular matrix-producing fibroblasts to model or support vascularized tissue. In a biomaterial scaffold or hydrogel, endothelial cells organize into vessel-like networks, while fibroblasts deposit matrix proteins and release paracrine signals that regulate endothelial adhesion, migration, and maturation. These co-culture systems help recreate cell-cell and cell-matrix interactions that are difficult to study in conventional two-dimensional cultures. In bioengineering, they support research on vascular development, wound healing, tissue regeneration, disease modeling, and therapeutic screening, while providing platforms for evaluating how biomaterials and culture conditions influence vascular organization.

Several protocols have been developed and described for the isolation of different cardiac cell types from a rat heart. Here, an optimized protocol is described that allows the isolation of high-quality major cardiac cell types (cardiomyocytes, endothelial cells, and fibroblasts) from a single preparation, reducing the experimental costs.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Fibroblast-Derived 3D Matrix System Applicable to Endothelial Tube Formation Assay

The aim of this method is to obtain fibroblast-derived 3D matrices as a natural scaffold for subsequent cellular assays. Fibroblasts are seeded in a pre-treated culture plate and stimulated with ascorbic acid for matrix generation. Matrices are decellularized and blocked to culture relevant cells (e.g., endothelial cells).

Microfluidic flow chambers etched by photolithography and fabricated from PDMS are applied to probe functional outcomes associated with EC dysfunction and inflammation. In a representative experiment, the ability of differential shear stress to modulate monocytic cell adhesion to cytokine activated EC monolayers is demonstrated.
