Suspension Culture Technique

Suspension culture technique is a method for growing cells, microorganisms, or cell aggregates throughout a liquid growth medium rather than attached to a solid surface, enabling controlled production in bioengineering. Gentle agitation keeps the culture mixed, improves access to nutrients and oxygen, and limits sedimentation, while temperature, pH, aeration, and mixing conditions are adjusted to support viable, uniform growth. This approach supports cell expansion and production of biological materials in vessels that can scale from laboratory flasks to bioreactors, making it useful for tissue engineering, biopharmaceutical manufacturing, and studies of cell growth under defined conditions.

A novel protocol for the mechanical preparation of testicular cell suspensions from rodent material, avoiding enzymes and detergents, is described. The method is very simple, fast, reproducible, and renders good quality cell suspensions, which are suitable for flow sorting and RNA extraction.

Generating an Organoid Culture from a Brain Tumor Cell Suspension

2025

This video describes the preparation and cultivation of brain tumor organoids from single-cell suspension. Using a laminin-rich extracellular matrix as a scaffold and culture media for nutrients, cells are facilitated to multiply and self-organize within the organoid. Consequently, ex vivo organoids are generated, mimicking cellular diversity and the microenvironment of brain tumors.

The tail-suspension test is validated as an experimental procedure to assess antidepressant efficacy of drug treatments in mice. Mice are suspended by their tails for six minutes and escape-related behaviors are assessed. We describe procedures used in conducting the tail suspension test.

A Technique to Culture Mouse Hippocampal Neurons

2025

The video demonstrates a method for culturing neurons from murine hippocampi. Initially, the hippocampi undergo enzymatic digestion to break down the extracellular tissue matrix. Subsequently, the digested tissue is mechanically disrupted to isolate individual neurons. Finally, these neurons are cultured on glial feeder cells.
